Pretreatment

After de-waxing the tissue slices they are treated with 0.2% hyaluronidase (app. 300 U/mg e.g. Art. No. HYA02-50) in TBS 15 min at 37°C. Thereafter non-specific binding is blocked by blocking serum or 3% BSA in TBS. For peroxidase systems blocking with 1% peroxide solution in TBS for 30 min at RT is recommended.

Anti-rabbit IgG-conjugates, e.g. anti-rabbit IgG:FITC (Art. No. FI-1000) or anti-rabbit IgG:DyLight488 (Art. No. DI-1488). For paraffin sections best results have been obtained by use of DAKO Envision or VECTOR ImmPress systems (Art. No. MP-7401).

This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided.

Properties

These will need colder storage temperatures.Polyclonals can be used for Western blot, immunohistochemistry on frozen slices or parrafin fixed tissues. The advantage is that there are more epitopes available in a polyclonal antiserum to detect the proteins than in monoclonal sera.Rabbits are used for polyclonal antibody production by bio logo. Rabbit antibodies are very stable and can be stored for several days at room temperature. bio logo adds sodium azide and glycerol to enhance the stability of the rabbit polyclonal antibodies.
