Secondary Antibodies

Long description

The antiserum is reacting with the Fab subunits IgG, IgA, IgM and other Ig classes of both light chain type, with their Fab or F(ab’)2 subunits and with free light chains of kappa and lambda type. It does not react with any non-Ig protein in chicken serum, as tested by immunoelectro- phoresis and double radial immunodiffusion. In precipitating techniques as immunoelectrophoresis and single and double radial immuno- diffusion (Mancini, Ouchterlony) to identify and measure immunoglobulins; as secondary antibody to precipitate the immunoglobulin in normal chicken serum.

Antibody come from

Purified Fab of normal IgG isolated from pooled chicken serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.

Other description

Delipidated, heat inactivated, lyophilized stable whole serum No preservative added, as it may interfere with the antibody activity. No foreign protein added. Total protein and IgG concentration in the antiserum are comparable to those of pooled goat serum. No foreign proteins added.

Antibody's reacts with these species

Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since homologous proteins of different species frequently share antigenic determinants. Cross- reactivity of this antiSerum has not been tested in detail.

Application

Immunoprecipitation

Antibody's suited for

Immunoprecipitation. In immunoelectrophoresis use 2 μl or equivalent against 120 μl antiserum. In double radial immunodiffusion use a rosette arrangement with 10 μl antiserum in a 3 mm diameter centre well and 2 μl serum samples (neat and diluted) in 2 mm diameter peripheral wells Precipitin titre not less than 1:64 when tested against normal chicken serum in agar block titration.

Storage

The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. It is reconstituted by adding 1 ml sterile distilled water. Working dilutions are prepared by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the product. Lyophilized at +4° C--at least 10 years reconstituted at or below -20° C--3-5 years reconstituted at +4° C--7 days

Description

This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided.

Properties

Chickens like all bird species have IgY antibodies that are very stable and found in the yolk. Gallus Gallus domestica is the Latin name of the Chicken.Human Fab Of Normal IgG Chromatographically purified Fab fragment of human IgG. Purified human Fab may be used as a reference antigen, calibrator, coating protein and blocking agent

Additional isotype

Latin name

About

Immunoglobulin gamma, IgG, mouse monoclonal H&L chain clones or rabbit, goat polyclonal antibodies have 4 parts. There are 2 heavy chains, 2 light chains. The IgG antibody has 2 antigen binding sites. They represent 70% or more of serum antibodies. This antibody can be antigen purified or protein A or G purified. For storage sodium azide is added or you can call us to request azide free antibody preparations. These will need colder storage temperatures.
